I'm hooking up my trailer to a septic tank (barrel). Should I remove the holding tank? Or let the stuff build up and pull the valve handle? I'd rather run it like a regular toilet, but the water flow is not high enough (so I've been told). Any advice?

I think you are supposed to let it build up and pull the valve handle.
